The first four episodes of the first season revolved primarily around characters discovering their powers, dealing with the issues of normal life and coping with the consequences of their discovery.
Hiro begins to discover that he can stop time and teleport and finds himself in New York city after testing his powers. Niki is having memory loss issues and finds herself waking up in strange places in different clothes and doesn't know what's going on. Claire is testing out her ability to heal from all injuries with the help of a friend. Matt is discovering his telepathy while on the path of a serial killer named Sylar who kills in strange, seemingly impossible ways. Isaac is freaking himself out by painting drawings which turn out to be true. Mohinder is trying to continue his father's research on people with genetic mutations which give them extraordinary powers. And Peter is having dreams about flying, and tries to convince his brother that he has super powers while his brother, Nathan, busies himself with running for Congressman.
At the end of the fourth episode, a smaller arc began with the message "Save the cheerleader, save the world", which is tied to the explosion already foreseen. Basically, Isaac paints a terrible explosion happening in the future and we're told that in order to prevent it the cheerleader (Claire) needs to be saved to save the world. This job falls to Peter who through clues in Isaacs paintings manages to find Claire and saves her from Sylar (who is the serial killer mentioned above. He is killing people with powers so that he can absorb them and become even more powerful). In doing this, Peter realizes he was absorb people's powers by coming into close contact with them, which will become quite an asset.
During this time, we learn that HRG is working for people that are trying to capture and study people with abilities, and that HRG knows what Claire is, but is willing to do anything to keep her out of the hands of the people he works for.
Niki is losing herself more and more to the Jessica personality, and eventually Jessica basically takes over and goes on a bit of a crime spree working for a crime boss named Linderman (who also has an interest in the people with abilities).
Eventually we learn that Mohinder's father was able to create a list of people of a genetic marker that indicated they would have special abilities. Sylar was one of the first ones he interviewed and he stole the information and commenced to start killing the people on the list to steal their abilities.
We find out that Linderman actually has abilities too, and was once part of an older generation of "specials" who fought for the greater good until greed and corruption ruined their team and they went their separate ways. Since then he's been manipulating others (he set D.L. and Niki up to marry and have a superpowered baby), and that he and Nathan and Peter's mother, have been setting up Nathan to become President of the United States of America for their own larger plan.
We learn that Claire is actually Nathan's daughter, and she get caught up in the Petrelli family drama before finally being reunited with HRG.
At the end of the season, Niki/Jessica (now with Niki mostly in control) tracks down Linderman and with the help of an injured D.L. manages to kill him. Peter, now suped up with many others powers, takes on Sylar and seemingly kills him (not really though) but Peter has absorbed the nuclear radiation Sylar was going to use to explode New York, and Nathan flies them both off after the fight (we don't know where). And Hiro, finds himself transported back to ancient Japan, where he will start off in the next season.
There are other little things in there, minor characters and whatnot, but I think that gives you a pretty good overview of how the characters started off, how they ended off and a little in the middle, lol
